Abstract:
Gaining the ability to reconstruct complex tissues would revolutionize medicine. I propose to achieve this ambition by reverse-engineering the tissues, which requires not only identifying individual mature cell types, but also unraveling where and how they have arisen from and developed into specific cell types.  We demonstrate the feasibility of reverse-engineering complex tissues via single-cell mapping and genome-wide molecular profiling of cell lineages in the adult Drosophila brain. We will extend further sophisticated tools to reverse-engineer higher brains in similar manners.  Knowing all the parts and learning how to manufacture and assemble them into a functional brain would help cure neurological disorders.
